高通量(high-throughput)生物實驗技術揭開後基因體世代 (post-genomic era)的序幕,而近年呈現爆炸性發展的次世代定序生物技術,更進一步將生物實驗技術推向高效率、高精度、高準確性之境界,為生命科學研究領域創造極佳的契機。次世代定序方法目前也已被廣泛應用於各類型的生命科學研究工作上,研究人員可針對DNARNA進行高速定序,搭配後端生物資訊 (bioinformatics)的應用,進行基因序列變異分析、基因結構異常分析、基因表現量分析、微小RNA (small RNA)表現量分析、基因體甲基化修飾等之研究。

本實驗室的研究主要在建立和應用各種NGS技術平台於癌症基因體及轉錄體上的研究搭配生物資訊的分析, 系統性的找出癌症重要的“生物特徵”相關病理特徵近年主要研究包括:

1.  口腔鱗狀細胞癌的全外顯組及轉錄組測序分析

2.  大腸直腸癌的外顯組轉錄組測序分析

3.  鼻咽癌靶向基因組重測序分析

4.  EV71感染宿主反應轉錄組測序分析

5.  建立口腔癌動物模型並對其癌症組織進行轉錄組測序分析

6.  從急性腎損傷和慢性腎臟疾病尿液樣本粹取小RNA並進行小RNA定序分析

7.  建立RNA-IP並利用序法找出RNA結合蛋白的標的RNA目標

此外我們更利用細胞和動物模式研究來驗証這些癌症相關標的,系統性的描繪這些​​變體的病理作用的基本分子機制。我們的最終目標是要揭示序列變或是新的診斷或治療靶基因調控機制。
