研究方向研究室特色

蛋白質體學研究是指全面性的探討蛋白質表現程度的學門。在生物醫學領域中,通常著重於找出兩個樣品之間相對差異表現的蛋白。由於分析技術的快速進步,使得蛋白質體分析的覆蓋率,已逐漸趕上基因體學研究。其中最主要的兩項技術為質譜學及胜肽分離技術。本實驗室主要的研究興趣就是針對這兩項技術進行研發及改進,以提升蛋白質體數據的廣泛度、靈敏度、可信度及自動化程度。
對於肽分離系統的改進,我們建立了多種奈米級多維液相分離系統,不論是一般性的整體蛋白體分析,或是特定族群胜肽的純化分離,其效能均已達國際一流的水準。利用這些系統,我們可以從30ug的細胞蛋白萃取物中鑑定出6000-9000個蛋白,甚至可從200微克蛋白中鑑定出近萬條磷酸化肽。至於在質譜測量方面,我們著重於開發嶄新的同位素標定技術,以提高質譜定量數據的可靠性。目前我們已建立兩種標定技術,其中之可進行整體蛋白體定量分析,另一則專注的定量特定胺基酸的氧化程度。這些技術都將運用於疾病蛋白體學研究,尤其將著重於分析臨床的組織或體液樣品。
