高通量(high-throughput)生物實驗技術揭開後基因體世代 (post-genomic era)的序幕,而近年呈現爆炸性發展的次世代定序生物技術,更進一步將生物實驗技術推向高效率、高精度、高準確性之境界,為生命科學研究領域創造極佳的契機。次世代定序方法目前也已被廣泛應用於各類型的生命科學研究工作上,研究人員可針對DNARNA進行高速定序,搭配後端生物資訊 (bioinformatics)的應用,進行基因序列變異分析、基因結構異常分析、基因表現量分析、微小RNA (small RNA)表現量分析、基因體甲基化修飾等之研究。

本實驗室的研究主要在建立和應用各種NGS技術平台於癌症基因體及轉錄體上的研究搭配生物資訊的分析, 系統性的找出癌症重要的“生物特徵”相關病理特徵,利用大數據庫運算,建構其可能參與之調控網絡,並利用細胞和動物模式進行深入的分子機制探討及驗証。近年主要研究包括:

  1. 建立台灣本土癌症組織之功能性基因體體學數據庫

建立完整的台灣本土癌症組織(口腔癌/食道癌/大腸直腸癌/腦癌)之功能性基因體體學數據庫,並利用生物資訊方式,結合國外大數據庫系統性的分析找出與臨床的早期診斷、疾病的治療與用藥、抗藥性、存活率與再復性相關的重要分子,完整的描繪這些變體的病理作用機制以及調控網絡。針對其中重要的變異基因,進一步使用細胞和動物模式研究來深入探討及驗証這些癌症相關標的之分子機制。

  1. 開發分析非編碼RNA調控網絡之生物資訊工具

利用非編碼RNA已知之生物特性 (long noncoding RNA/circRNA/miRNA),結合大數據庫,開發分析非編碼RNA調控網路之生物資訊分析平台。進一步結合細胞和動物模式,驗証其中新穎之非編碼RNA的調控分子機制。

  1. 開發次世代定序法於癌症液態檢體之應用

外泌體組成不僅反應了起源細胞,且富含與細胞轉化相關的生物分子,因此已成為近年來液體活檢方法中最有潛力的目標。實驗室建構了一套只需1 cc的血漿,即可快速提取外泌核糖核酸方法,利用次世代定序法驗証了此方法明顯優化並提升樣本製備的穩定性。我們更進一步全面性的建立了健康人及多種癌症之外泌微核糖核酸體學數據庫 (口腔癌、食道癌、肺癌、大腸直腸癌),結合臨床資訊,系統性的尋找具早檢測、長期監測治療反應以及預後之多面向的生物標誌。因應精準醫學方面之應用,更進一步開發一套快速檢測晶片系統,可精準且多面向的監測癌症及治療進程。利用多種生物資訊工具,完整描繪出外泌微核糖核酸在癌症進程所扮演的功能角色,並在細胞及動物模式上進行深入的機制研究。
