研究方向及研究室特色: (1) 了解泛素化(ubiquitination)對於發育及癌細胞生長的角色 Ubiquitination屬於蛋白質後轉譯(post-translational) 調節的機制,主要由ubiquitin ligase決定那些蛋白受此機制調控。我們實驗室主要研究Cbl (E3 ligase)調節上皮生長激素(EGFR),對果蠅體軸發育及癌細胞生長造成的影響。果蠅卵發育中Cbl 藉由影響EGFR的內噬及分解作用,而調降EGFR訊息傳遞,造成體軸的改變。人類癌細胞中Cbl也藉由此機制影響EGFR 訊息傳遞而抑制細胞生長。近期,我們發現果蠅Cbl影響CTP合成酶的聚合構造(CTP synthase filament),而調節DNA的複製。Cbl 如何調解CTP合成酶的聚合構造,與此構造和癌症的關係是我們近期的研究方向。 (2) 代謝對果蠅再生與卵發育的影響 生物體常以調節代謝對外在環境作出快速反應。我們利用果蠅系統,了解蛋氨酸(Methionine)代謝促進果蠅卵發育,並藉由調節膜蛋白運輸而影響呼吸管的型態。同時,我們發現蛋氨酸(Methionine)代謝對果蠅翅膀再生產生影響。 蛋氨酸(Methionine)代謝如何影響蛋白運輸,及如何影響果蠅翅膀再生是我們近期的研究方向。
